DESCRIPTION OF A NEW ISOLATE OF CUCUMBER MOSAIC VIRUS IN IRAQ

This study was initiated to isolate and characterize Cucumber mosaic virus based on serological, biological, molecular approaches. Leaf samples were collected from symptomatic cucumber plants grown in protected fields at Plant protection Dept., College of Agricultural Engineering Sciences, the University Baghdad Al-Jhdryaa used for biological assays. ELISA using CMV specific commercial kit detect samples. RT-PCR primer set targeting CP gene confirm infection. DNA fragments amplified sequenced analyzed computer software packages. Bioassays showed Vigna unguiculata exhibited necrotic local lesions when inoculated with indicated that sample infected scoring 2.907 highest absorbance 405 nm. CMV-F/ CMV-R could amplify targeted ~500 bp only. Sequence analyses confirmed detection isolated shared 95 91% maximum nucleotide (nt) deduced amino acid (aa) sequence identities equivalent bank sequences India, Japan, China, USA. Neighbor-joining (NJ) phylogenetic tree, concentrated partial nt aa sequence, relatedness gouged other suggesting a common origin. These findings culture infected.
